Primary and Secondary Standards

Quantitative Analysis and Measurement

Analytical Methods

  • Stoichiometric Methods: Patterns are associated with chemical types that do not contain the analyte.
  • Comparative Methods: Require calibration of the measuring instrument's response relative to the concentration of the component of interest using chemical standards.

Reference Materials

  • Reference Material: A substance with properties sufficiently well-established to be used for instrument calibration, estimation of a measurement method, or assigning values to materials.
  • Certified Reference Material (CRM): A material where property values are certified by a technically valid procedure, accompanied by a certificate or official documentation.
  • Standard Reference Materials: Materials with specific certified physical or chemical properties.

Calibration Principles

Calibration involves checking the instrument's response against a pattern of known properties to ensure accuracy.

Types of Calibration

  1. Instrumental Calibration: Ensures the proper functioning of the instrument used for quantization (e.g., balance, burette).
  2. Methodological Calibration: Establishes an unequivocal relationship between the instrumental response and the concentration of the analyte.

Methodological Calibration Techniques

  • Simple/External Standard Calibration: Compares sample signals with a set of signals from individual calibration samples. The analytical process is applied to both calibration and test samples independently.
  • Standard Addition: Recommended when ensuring similarity between the behavior of patterns and the sample matrix is required. This method standardizes the analysis within the sample matrix itself.
  • Internal Standard: Minimizes instrumental fluctuations and provides references for qualitative and quantitative purposes. A known quantity of a compound not present in the sample matrix is added to both the standard and the test sample. The internal standard must be compatible with the sample and possess physical and chemical characteristics similar to the analyte.
